The application of this reaction in intramolecular cyclization is common in natural product synthesis. This selectivity in the formation of a C–C bond was first discovered by Giese and has found several applications in organic synthesis. 1 The reaction is very efficient when alkyl radicals (high‐lying SOMO) are added to electron‐deficient olefins (low‐lying LUMO), becoming a good alternative to the Michael reaction. 2 This strategy Tin-free Giese reaction of alkyl iodides with electron-deficient alkenes and the related radical carbonylation process proceeded efficiently in the presence of sodium cyanoborohydride and tetrabutylammonium cyanoborohydride. Transfer of iodine followed by hydride reduction of the resulting carbon-iodine bond is proposed as a possible mechanism. With the use of an automated flow microreactor, flow reaction conditions for the Giese reaction were quickly optimized, and it was found that a reaction temperature of 70 °C in combination with a residence time of 10-15 minutes gave good yields of the desired addition products. Tin-free Giese reactions, employing primary, secondary, and tertiary alkyl iodides as radical precursors, ethyl acrylate as a radical trap, and sodium cyanoborohydride as a radical mediator, were examined in a continuous flow system.

For the generation of the initial radical a broad variety of suitable precursors can be employed, such as halides, thio‐ and selenoethers, alcohols, aldehydes and hydrocarbons. Recent advances in photoredox catalysis have demonstrated the generation of carbon-centered radicals under mild conditions, 4 which has substantially filled the existing methodology gaps on conjugate addition reactions to electron-deficient olefins, also known as the Giese reaction. 5 Several research groups have recently reported on the generation of carbon-centered radicals from a diverse set of … Scheme 1: Giese reaction using borohydride-based radical mediators. In recent years, microreaction technologies have made a significant impact on chemical synthesis and production in terms of their advantageous characteristics, which include efficient mixing, efficient mass and heat transfer, and high operational safety [19-23].

In general, radical cyclization reactions comprise three basic steps: selective radical generation, radical cyclization, and conversion of the cyclized radical to the product. For the generation of the initial radical a broad variety of suitable precursors can be employed, such as halides, thio‐ and selenoethers, alcohols, aldehydes and hydrocarbons.

An efficient cobalt-catalyzed difluoroalkylation/Giese radical conjugate cyclization manifold with various alkyne-tethered cyclohexadienones and halogenated fluorinating reagents was accomplished under remarkable mild reaction conditions, thus providing an expedient route to valuable fluorinated chromens. Introduction. Radical cyclization reactions produce mono- or polycyclic products through the action of radical intermediates. Because they are intramolecular transformations, they are often very rapid and selective.
